I think lovely Tisiphone starts to learn new word when you meet her without upgraded bosses in the Pact.
Also armorless Theseus and Asterius got new attack move after subplot when Theseus thought that Zag tries to quarrel him with Asterius.
To play lyre better you just need to practice on it more and more, like you would in reality. I can confirm having progress in between runs right there on the spot by playing on it repeatedly. When i first bought it, i thought it would be like that board game, or Hercules weights – always same result, but for whatever reason i kept pressing on it, then gave up, than continued again, and one day – progress. From regular "trangs" and "broons" to actual string notes, then chords, then sweet tunes. It actually inspired me to start learning how to play piano. I found an old simple synthesizer in some basement and decided to give it a try, now i can play bits from "Entertainer" and "In the Hall of the Mountain King". =)
