The Ancient Greek-themed action adventure was the big winner of the night, taking away five awards in total.
The title, made for PC and Nintendo Switch, has been commended by critics for its unique art style and distinctive gameplay.
Fans voted voted The Last of Us Part 2 as the EE Game of the Year.
Sackboy: A Big Adventure took the British Game, Animal Crossing: New Horizons won in the Multiplayer category and the Game Design award went to Hades.
It was the second time the awards ceremony was held online due to lockdown.
Completing Hades in the middle of a pandemic had its challenges but it’s something Greg Kasavin, creative director at Supergiant Games, will never forget.
“The trust that we put in each other – both was tested in some respects but was really just it’s what got us through into this,” he tells Radio 1 Newsbeat.
“It’s something to never, ever take for granted – that as a group, we all believe in each other in this way.”
Studio director and co-founder Amir Rao agrees: “Hades was a game that a lot of people played for a long time, and hopefully it helped them in some way, and they came out with a lasting positive impression as a result of their time with it.”

At least five people have been killed in Alabama, as tornadoes continue to batter the southern US state.
“Tragically, we are receiving reports of loss of life,” Governor Kay Ivey said. She urged residents to be on high alert.
The five deaths were in the small town of Oatchee in Calhoun County, the local emergency agency said, as the storm damaged houses and uprooted trees.
The tornadoes later hit Chilton County and were nearing Shelby Country.
“DO NOT WAIT TO SEE THE TORNADO! If you are in its path, TAKE SHELTER NOW!” the National Weather Service tweeted.
Social media users posted dramatic footage of the fast-moving tornadoes wreaking havoc in Alabama.
In 2019, more than 20 people – including three children – died after two tornadoes caused extensive damage to buildings and roads in eastern Alabama.
First Lady Jill Biden has cancelled a planned trip to the state with actress Jennifer Garner due to the storms. Mrs Biden had been due to be in Birmingham to discuss the benefits of the newly-passed Covid relief package.

The award-winning US actress Jessica Walter has died aged 80, her family says.
“It is with a heavy heart that I confirm the passing of my beloved mom,” Walter’s daughter Brooke Bowman said.
“A working actor for over six decades, her greatest pleasure was bringing joy to others through her storytelling both on screen and off,” Ms Bowman said.
Walter is best known for playing icy matriarch Lucille Bluth in the Arrested Development TV sitcom in the 2000s.
She also won an Emmy for starring as a detective in the police drama Amy Prentiss in the 1970s.
“While her legacy will live on through her body of work, she will also be remembered by many for her wit, class and overall joie de vivre,” Ms Bowman said.
The Deadline website, which first reported the news, quoted the family as saying that Walter had died in her sleep at her home in New York.
Some of Walter’s most memorable lines in Arrested Development were delivered over a Vodka Martini or between glasses of wine.
During her long career, Walter starred in several popular TV dramas and sitcoms, like NCIS and Law & Order, and also appeared in a number of films.
She also voiced Malory Archer in the animated series Archer.
One of Walter’s starring movie performances was alongside Clint Eastwood in his 1971 directorial debut, Play Misty for Me, earning a Golden Globe nomination.
She also starred in Grand Prix (1966).
Among her TV comedy cameos, Walter played Mrs Latham, a university benefactor who seduces Leonard in an episode of The Big Bang Theory.
Tony Hale, who played Walter’s son in Arrested Development, paid tribute to her, saying “her talent and timing were unmatched”.
US actor and stand-up comedian David Cross described Walter as “an absolutely brilliant actress and amazing talent”.
“I consider myself privileged and very lucky to have been able to work with her. Lucille Bluth is one of TV’s greatest characters,” Cross wrote in a Twitter post.
